Until now, pyridoxine (PN), the most common vitamer B6 comes to animals and humans, are chemically synthesized for commercial purposes. Thus, the development of microbial fermentation process is very attractive to the biotechnology industry.
More recently, we construct strain Bacillus subtilis were formed in a significant number of PN by non-native deoxyxylulose 5′-phosphate- (DXP) -dependent pathway of vitamin B6. Here we report on the optimization of the condensation reaction of this pathway consisting of 4-hydroxy-L-threonine-phosphate dehydrogenase pDXA, which pyridoxine 5′-phosphate synthase original PdxJ and DXP synthase, DXS. To enable eat high amounts of 4-hydroxy-threonine (4-HO-Thr) which can be converted to a PN by B. subtilis overexpressing pDXA and PdxJ, we first adjusted the bacteria to tolerate antimetabolite 4-HO-Thr. Adapted bacteria produced 28-34mg / l PN of 4-HO-Thr whereas the wild-type parent only produced 12 mg / l PN.
Moreover, by expressing alleles differ in their pdxJ pDXA and adapted strains we identified a better combination of enzymes pDXA and PdxJ than previously reported, and the resulting strain produced 65 mg / l PN. To further improve productivity mutants were isolated that efficiently retrieve and convert deoxyxylulose (DX) to DXP, which is inserted into the PN. Although the mutant highly efficient for converting low amounts of exogenous DX, at a higher level of DX they performed only slightly better. This study found some enzymes with promiscuous activities and revealed that the host’s metabolic pathways compete with heterologous pathway for 4-HO-Thr. In addition, the study showed that B. subtilis genome is quite flexible with respect to adaptive mutation, the property, which is very important for strain engineering.
Qualitative tests compared with the gold standard quantitative test. G6PDd higher prevalence of RDT (8.6%) compared with the quantitative assay (6.8%), using <60% threshold of activity. RDT performed optimally at <threshold of 60% and showed high sensitivity (≥ 90%) and negative predictive values (100%) in the three threshold of activity (under 60%, 30% and 40%).
G202A allele frequency was 6.4%, 7.9%, and 6.8% among women, men, and overall, respectively. Notably, more than half of G202A homo / child hemizygous states ≥ 60% activity of the enzyme. Overall, CareStart ™ G6PD RDT seems to be a feasible screening test to accurately identify individuals with enzyme activity below 60%. G6PDd low prevalence in all three diagnostic modalities and the absence of severe deficiency in our study suggests that there is little resistance to the use of a single dose of primaquine in this region.
Optimization of DNA amplification hexaplex of short tandem repeat loci amelogenin.
An automated system of DNA profiling based multiplex amplification of highly polymorphic short tandem Repeat (STR) loci amelogenin markers and developed. Five STR loci with overlapping allele size range has been used in the multiplex amplification, including HUMD1S103, HUMTH01, HUMD21S11, HUMD18S51, and HUMFIBRA.

Description: CRK, also known as p38, is a protein that in humans is encoded by the CRK gene. This gene is a member of an adapter protein family that binds to several tyrosine-phosphorylated proteins. It is mapped to 17p13.3. The protein participates in the Reelin signaling cascade downstream of DAB1. The product of this gene has several SH2 and SH3 domains (src-homology domains) and is involved in several signaling pathways, recruiting cytoplasmic proteins in the vicinity of tyrosine kinase through SH2-phosphotyrosine interaction. The N-terminal SH2 domain of Crk functions as a positive regulator of transformation whereas the C-terminal SH3 domain functions as a negative regulator of transformation. Two alternative transcripts encoding different isoforms with distinct biological activity have been described. |
