BACKGROUND
Studies on the impact of the load on the allele JAK2V617F and clinical phenotypic traits in Philadelphia-negative myeloproliferative neoplasms (MPNs Ph-negative) have reported variable results. We aimed to analyze the association JAK2V617F mutated allele burden with the characteristics of laboratory and clinical phenotype in patients Turkey (107 essential thrombocythemia (ET) and 77 primary myelofibrosis (PMF)).
METHOD
Peripheral blood samples from 184 patients with Ph-negative MPNs analyzed for JAK2V617F allele status and load. MutaScreen JAK2 assay (Ipsogen, Luminy Biotech, Marseille, France) was used to detect the status JAK2V617F and JAK2V617F quantitative allele burden in genomic DNA using the TaqMan allelic discrimination.
RESULTS
Frequency JAK2V617F-positive patients with a high mutation load (allele burden >> 50%) higher in comparison with ET PMF (23.4% and 4.7%, respectively; P = 0.001). We found a significant association between patients ET with JAK2V617F high-allele burden and low hemoglobin (Hb) and hematocrit (Ht), LDH levels were higher and massive splenomegaly more common (P = 0.001, P = 0.001, P = 0.012 and P = 0.015, respectively). ET patients with a high mutation load shown a higher prevalence of bleeding compared with a low mutation load and the wild-type mutation status (P = 0.003). DVT level was significantly higher in patients with ET with a mutant allele burden at the top compared to reduce by half and the wild-type (P = 0.029). We observed a significant association between patients PMF with high JAK2V617F allele burden and higher hemoglobin, hematocrit level and the number of leukocytes (P = 0.003, P = 0.021 and P = 0.001, respectively).
CONCLUSION
Our study shows JAK2V617F load allele correlated with clinical features in ET and PMF. We conclude JAK2V617F quantification of mutations contributing to the examination of Ph-negative MPNs.
[The relationship between vitamin D receptor gene polymorphism and chronic periodontitis].
OBJECTIVE
To investigate the relationship between vitamin D receptor (VDR) gene polymorphism and chronic periodontitis (CP).
METHOD
buccal swabs of 105 patients with mild CP / mode-level and 85 severe CP collected, this DNA was extracted from buccal swabs using DNA Swab Kit TIANamp [TIANGEN Biotech (Beijing) CO.Ltd]. VDR rs1544410 and rs731236 were genotyped by Sequenom MassARRAY system (Shanghai Benegene Biotechnology Co. Ltd.), which is based on MALDI-TOF technology (matrix-assisted laser desorption ionization time-of-flight). Genotype distribution and allele frequency was analyzed.
RESULTS
Frequency of rs1544410 A allele and AA + AG genotype were significantly higher in CP severe than in mild / moderate CP of all patients and female patients respectively (all patients: P = 0.006, 0.007, whereas female patients: P = 0.001, 0.001 ). Frequency of rs731236 C allele and CC + CT genotype was significantly higher in CP severe than in mild / moderate CP of all patients and female patients respectively (all patients: P = 0.003, 0.004, whereas female patients: P <0.001, < 0.001).
CONCLUSION
VDR gene polymorphism rs731236 and rs1544410 may be associated with severe CP in the Han Chinese population.

Description: CRK, also known as p38, is a protein that in humans is encoded by the CRK gene. This gene is a member of an adapter protein family that binds to several tyrosine-phosphorylated proteins. It is mapped to 17p13.3. The protein participates in the Reelin signaling cascade downstream of DAB1. The product of this gene has several SH2 and SH3 domains (src-homology domains) and is involved in several signaling pathways, recruiting cytoplasmic proteins in the vicinity of tyrosine kinase through SH2-phosphotyrosine interaction. The N-terminal SH2 domain of Crk functions as a positive regulator of transformation whereas the C-terminal SH3 domain functions as a negative regulator of transformation. Two alternative transcripts encoding different isoforms with distinct biological activity have been described. |
